(4-Aminobutyl) guanidine - Protective effect of Agmatine on vascular endothelial injury in septic mice

Abstract:

Objective: to explore the effect of Agmatine on vascular endothelial function in septic mice. Methods: A mouse model of sepsis was established by cecal ligation and puncture (CLP). Forty-two 6-8 week-old male C57BL/6 mice were randomly divided into sham operation group (14 mice, intraperitoneal injection of phosphate buffer),CLP model Group (14 mice, intraperitoneal injection of phosphate buffer), in the agmatine-treated group (14 rats, intraperitoneal injection of 400mg/kg agmatine), mice in each group were sacrificed 24h after modeling and blood samples were collected, and liver and lung tissue samples were perfused. TNF-α, interleukin (IL-6,IL-10), intercellular adhesion molecule -1(ICAM-1) in serum were detected by enzyme-linked immunosorbent assay (ELISA), vascular adhesion molecule -1(VCAM-1), Vascular endothelial growth factor (VEGF), hemophilia coagulation factor (vWF), angiotensin -2(Ang-2), chemokine (MCP-1). The fluorescence intensity of liver and lung tissue lysates was detected by FITC-dextran fluorescence tracer method. Results: 24h after modeling, the abdomen of the CLP Group was light pink, the body temperature decreased, the spirit was poor, and the activity was reduced. After agmatine treatment, the spirit and activity of the mice were significantly improved. Compared with sham operation group, the content of vascular endothelial biomarkers in serum and plasma of CLP Group was significantly increased, and agmatine treatment could significantly inhibit its content (P0.01). At the same time, the fluorescence intensity of liver and lung tissue in CLP Group was significantly higher than that in sham operation group (P0.01), while agmatine could significantly reduce the fluorescence intensity of liver and lung tissue (P0.01). In addition, compared with sham surgery, In CLP Group, the levels of serum inflammatory factors were significantly increased (P0.01), while the expression of inflammatory mediators was significantly decreased after agmatine treatment (P0.01). Conclusion: agmatine can reduce the vascular permeability of liver and lung tissue by inhibiting the secretion of endothelial related molecules and inflammatory factors such as endothelial adhesion molecules, vascular endothelial growth factor and hemophilia coagulation factor, so as to alleviate the vascular endothelial injury in septic mice.

Abstract:

Objective: to observe the relationship between the anti-physical dependence effect of agmatine and opioid receptor in rats. Methods: The experiment was carried out by two methods: Rat physical dependence experiment and radioligand binding experiment. Results: Although agmatine could inhibit the withdrawal reaction of morphine dependent rats induced by naloxone in a dose-dependent manner, however, it has no inhibitory effect (no direct effect) on the binding reaction of 3h-naloxone to opioid receptors in rat forebrain, neither does it affect compensatory adaptive changes (without indirect effects) such as decreased affinity of opioid receptors for ligands and down-regulation of receptor numbers when physical dependence occurs. Conclusion: Although agmatine can inhibit opioid dependence, it has no direct or indirect effect on opioid receptors.
